運動論文:不同運動訓(xùn)練時間大鼠心肌t-mTOR和p-mTOR蛋白的表達(dá)【中文摘要】:通過大鼠運動訓(xùn)練后心肌t-mTOR和p-mTOR蛋白表達(dá)的變化,來探討它們在運動誘發(fā)心肌功能變化過程中的作用以及運動對mTOR信號通路的影響。方法:急性組運動模型:選用雄性清潔級SD大鼠50只,隨即分為安靜組11只和一天急性訓(xùn)練組(運動后即刻13只、運動后3小時13只、運動后24小時13只),安靜組不運動,只進(jìn)行正常的籠內(nèi)生活,大鼠適應(yīng)實驗環(huán)境3天后開始訓(xùn)練,運動組參照Bedford漸增負(fù)荷跑臺運動模型訓(xùn)練。一個月組運動模型:雄性清潔級SD大鼠50只,隨即分為安靜組11只和一個月訓(xùn)練組(運動后即刻13只、運動后3小時13只、運動后24小時13只),安靜組不運動,只進(jìn)行正常的籠內(nèi)生活,大鼠適應(yīng)實驗環(huán)境3天后開始訓(xùn)練,參照Bedford方法和王蘊(yùn)紅的長期遞增負(fù)荷耐力訓(xùn)練建立運動模型,采用跑臺方式訓(xùn)練。三個月組運動模型:雄性清潔級SD大鼠50只,大鼠適應(yīng)環(huán)境后隨機(jī)分為安靜組11只和耐力訓(xùn)練組(運動后即刻13只、運動后3小時13只、運動后24小時13只),大鼠適應(yīng)實驗環(huán)境3天后開始訓(xùn)練,參照Bedford方法和王蘊(yùn)紅的長期遞增負(fù)荷耐力訓(xùn)練建立運動模型,采用跑臺方式訓(xùn)練。大鼠每周訓(xùn)練六天,周日休息,共訓(xùn)練10周,具體的訓(xùn)練方案是:安靜對照組大鼠為正?;\內(nèi)飼養(yǎng),不進(jìn)行任何運動。運動組大鼠每周周一至周六運動,周日休息。每周一開始運動之前稱體重并記錄。末次運動后按實驗設(shè)計的各個時間點取材。取大鼠左心室心尖。然后收集蛋白樣品(研磨,提蛋白,測蛋白,調(diào)蛋白,煮蛋白,分裝),電泳,轉(zhuǎn)膜,封閉,一抗孵育,二抗孵育,蛋白檢測。所得數(shù)據(jù)用統(tǒng)計軟件分析。結(jié)果:急性一天訓(xùn)練組大鼠t-mTOR和p-mTOR在大鼠左心室心肌的表達(dá)安靜組與運動組表達(dá)沒有規(guī)律性變化。一個月訓(xùn)練組大鼠t-mTOR和p-mTOR在大鼠左心室心肌的表達(dá)情況為:運動后3小時,運動后24小時與安靜對照組相比表達(dá)升高,差異不顯著。三個月訓(xùn)練組大鼠t-mTOR和p-mTOR在大鼠左心室心肌的表達(dá)情況為:運動后3小時,運動后24小時與安靜對照組相比表達(dá)升高,差異顯著。結(jié)論:1急性運動后大鼠左心室心肌t-mTOR和p-mTOR蛋白表達(dá)沒有顯著變化。2一個月訓(xùn)練組大鼠t-mTOR和p-mTOR在大鼠左心室心肌的表達(dá)情況為:運動后3小時,運動后24小時與安靜對照組比表達(dá)升高但沒有顯著性差異。表明一個月訓(xùn)練后也沒有引起t-mTOR和p-mTOR蛋白的顯著變化。3三個月訓(xùn)練組大鼠t-mTOR和p-mTOR在大鼠左心室心肌的表達(dá)為:運動后3小時,運動后24小時與安靜對照組相比表達(dá)升高,差異顯著。運動恢復(fù)期蛋白合成增加。表明mTOR信號通路與心肌肥大密切相關(guān)。mTOR信號通路可能參與了心肌功能的調(diào)節(jié)和形態(tài)的變化。【英文摘要】: to explore the role of exercise in my ocardial function in the process, as well as the impace of the exercise,we observe the expression of the member of t- mTOR : Model of acute training:we selected 50 male clean SD rats for actute exercise and they were randomly divided into a control group and acute exercise (instantly after exercise, control ,3 hour, 24 hours after exercise), the rats control group just lived a normal life in cafe in cage and dont exercise, after 3 days adaption,the actute exercise training group awere trained on the treadmill which refered to the Bedford increasing load of a month exercise group: were randomly divided into a control group and Group of a month exerciseing (instantly after exercise, control,3 hour, 24 hours after exercise),in 50 SD male rats. the rats control group just lived a normal life in cafe in cage and dont exercise, after 3 days adaption,the exercise training groups were trained on the treadmill which refered to the Bedford and wangyunhong increasing load of 3 month exercise group: were randomly divided into a control group and Group of a month exerciseing (instantly after exercise, control,3 hour, 24 hours after exercise),in 50 SD male rats. the rats control group just lived a normal life in cafe in cage and dont exercise, after 3 days adaption,the exercise training groups were trained on the treadmill which refered to the Bedford and wangyunhong increasing load model. Then the collection protein sample (attrition, raises protein, measures protein, adjusts protein, boils protein, subpackage), the electrophoresis, transfers the membrane, the seal, a anti-foster, two anti-fosters, protein obtained data counts the software analysis with in acute training the expression of t-mTOR and p-mTOR does not have the change on month training group the expression of t-mTOR and p-mTOR on myocardial :the express increased in 3 hours,24 hours after exercise compared with control group. The difference is not months training group the expression of t- mTOR and p-mTOR on myocardial :the express increased in 3 hours, 24 hours after exercise compared with control group. The difference is :1 in acute training the expression of t-mTOR and p-mTOR does not have the regular change on myocardial. it indicated mTOR signal passage is not remarkable on myocardial in acute A month training group the express increased in 3 hours, 24 hours after exercise compared with control express increased in 3 hours, 24 hours after exercise compared with control group. The difference is not remarkable. A month training that did not cause the change of t-mTOR and p-mTOR 3 months training group the expression of t-mTOR and p-mTOR on myocardial :the express increased in 3 hours, 24 hours after exercise compared with control group.
