外源添加木質(zhì)素對農(nóng)田土壤微生物的影響

1、    外源添加木質(zhì)素對農(nóng)田土壤微生物的影響    孫敬文 尹晗 安雪暉 何世欽3 孫月 陳禹竹摘要 目的研究木質(zhì)素對土壤微生物的影響，為木質(zhì)素后期的資源化利用提供一定的基礎(chǔ)資料。方法采集農(nóng)田土壤進(jìn)行培養(yǎng)試驗，設(shè)置空白對照和木質(zhì)素處理組，培養(yǎng)結(jié)束后提取土壤dna，結(jié)合定量pcr和高通量測序等技術(shù)，研究木質(zhì)素對土壤微生物的影響，對微生物的群落組成及多樣性進(jìn)行分析。結(jié)果定量pcr結(jié)果表明，木質(zhì)素的加入減少了細(xì)菌16s rrna基因豐度，且顯著降低了真菌18s rrna基因豐度。高通量測序結(jié)果顯示，alpha多樣性分析上，細(xì)菌的空白對照多樣性指數(shù)均高于木質(zhì)素處

2、理，真菌則相反，但均未達(dá)顯著水平;pcoa分析表明，木質(zhì)素的加入對細(xì)菌和真菌群落均產(chǎn)生了顯著影響，不同處理間的群落結(jié)構(gòu)差異較大。在門水平上對微生物群落組成分析表明，木質(zhì)素的加入顯著提高了細(xì)菌中-變形菌、擬桿菌、厚壁菌的相對豐度，降低了酸桿菌、放線菌、-變形菌的相對豐度;真菌中明顯增加了擔(dān)子菌、被孢霉菌的相對豐度，而降低了子囊菌的相對豐度。結(jié)論木質(zhì)素的加入對土壤中細(xì)菌、真菌的數(shù)量及群落結(jié)構(gòu)均產(chǎn)生了一定影響?？傮w上，木質(zhì)素不利于土壤微生物群落的生長繁殖和代謝。由于對木質(zhì)素的利用能力不同，不同微生物呈現(xiàn)出了不同的效果。關(guān)鍵詞 木質(zhì)素;土壤微生物;定量pcr;高通量測序中圖分類號 s 154.3

9、deltaproteobacteria. for the fungi， the relative abundance of basidiomycota and mortierellomycota was increased significantly， but the relative abundance of ascomycota was decreased. conclusion the addition of lignin has a certain impact on the copy number of microorganisms and community structure of bacteria and fungi in the soil. on the whole， lignin is not conducive to the growth and metabolism of soil microbial communities. due to the different utilization capacity of lignin， some microorganisms show different effects.key words lignin;soil microorganism;q
