摘要:本研究旨在探究高脂飲食對小鼠攝食及情緒活動的影響,并考察柴胡疏肝散的干預(yù)作用。通過選取健康的雄性C57BL/6J小鼠,隨機分為正常飲食組和高脂飲食組兩個實驗組,每組10只。同時,將高脂飲食組分為干預(yù)組和對照組,干預(yù)組在高脂飲食條件下口服柴胡疏肝散,對照組僅口服等量的蒸餾水。實驗結(jié)果顯示,相較于正常飲食組,高脂飲食組小鼠的日攝食量顯著升高(P<0.05),而柴胡疏肝散干預(yù)組的日攝食量則有所降低(P<0.05)。同時,高脂飲食組小鼠在開放場地測試中呈現(xiàn)出更多的抑郁行為,而柴胡疏肝散干預(yù)組的抑郁行為顯著下降(P<0.05)。這表明高脂飲食可引起小鼠的攝食量增加及情緒紊亂,而柴胡疏肝散在一定程度上對高脂飲食所致的情緒異常有較好的干預(yù)作用。

Abstract:Thisstudyaimedtoinvestigatetheeffectsofhigh-fatdietonfoodintakeandemotionalactivitiesinmice,andtoexploretheinterventioneffectsofChaihuShuganPowder.HealthymaleC57BL/6Jmicewererandomlydividedintothecontrolgroupandthehigh-fatdietgroup,with10miceineachgroup.Thehigh-fatdietgroupwasfurtherdividedintotheinterventiongroupandthecontrolgroup.TheinterventiongroupwasorallyadministratedwithChaihuShuganPowder,whilethecontrolgroupreceivedanequalvolumeofdistilledwater.Theresultsshowedthatthedailyfoodintakeofthehigh-fatdietgroupwassignificantlyhigherthanthatofthecontrolgroup(P<0.05),whilethedailyfoodintakeoftheinterventiongroupwasdecreased(P<0.05).Moreover,thehigh-fatdietgroupdisplayedmoredepressivebehaviorsintheopenfieldtest,whilethedepressivebehaviorsintheinterventiongroupweresignificantlyreduced(P<0.05).Thesefindingssuggestthathigh-fatdietcanincreasefoodintakeandcauseemotionaldisordersinmice,whileChaihuShuganPowderhasagoodinterventioneffectonemotionalabnormalitiescausedbyhigh-fatdiet.

Furthermore,thehigh-fatdietgroupshowedmoredepressivebehaviorsintheopenfieldtest,indicatingthatthehigh-fatdietmaybeassociatedwithemotionalabnormalities.Thisfindingisalsoinlinewithpreviousstudiesthathaveshownthathigh-fatdietscancausedepressive-likesymptomsinrodents.

However,theadministrationofChaihuShuganPowdertotheinterventiongroupresultedinasignificantreductionindepressivebehaviors.ChaihuShuganPowderisaChineseherbalformulathathasbeentraditionallyusedtotreatliverandmooddisorders.TheresultsofthisstudysuggestthatChaihuShuganPowderhasagoodinterventioneffectonemotionalabnormalitiescausedbyhigh-fatdiet.

Overall,thisstudyhighlightsthepotentialofdiet-inducedobesityasamodelforinvestigatingtherelationshipbetweendietandemotionaldisorders.ItalsoprovidesevidencefortheeffectivenessofChaihuShuganPowderinmitigatingemotionalabnormalitiescausedbyhigh-fatdiet.FurtherresearchisneededtoelucidatethemechanismsunderlyingtheseeffectsandtoexplorethepotentialclinicalapplicationsofChaihuShuganPowderintreatingemotionaldisordersassociatedwithobesityInadditiontothefindingsmentionedabove,thisstudyhasfurtherimplicationsforunderstandingthecomplexrelationshipbetweendietandemotionaldisorders.Diet-inducedobesityhasincreasinglybeenrecognizedasariskfactorforarangeofmentalhealthproblems,includingdepression,anxiety,andcognitiveimpairment(1).

Thismaybedueinparttotheimpactofdietongutmicrobiota,whichhasbeenshowntoplayacrucialroleinregulatingmoodandbehavior.High-fatdiets,inparticular,havebeenfoundtodisruptthebalanceofgutbacteriaandleadtochroniclow-gradeinflammation,whichmaycontributetothedevelopmentofemotionalabnormalities(2).

TheuseofChaihuShuganPowderinthisstudyalsoraisesintriguingquestionsaboutthepotentialfortraditionalChinesemedicinetooffernewinsightsandtreatmentsforemotionaldisordersassociatedwithobesity.Althoughmanytraditionalremedieshavebeenusedforcenturiestotreatmentalhealthproblems,thereisstillmuchtolearnabouttheirmechanismsofactionandpotentialclinicalapplications.

FutureresearchinthisareacouldinvolveexaminingthespecificcompoundsandpathwaysinvolvedintheeffectsofChaihuShuganPowderonemotionalregulation,aswellasexploringthebroaderpotentialfortraditionalremediesinaddressingthecomplexinterplaybetweendiet,guthealth,andmentalhealth.

Overall,thisstudyaddstoagrowingbodyofevidencesuggestingthatdiet-inducedobesityisasignificantriskfactorforemotionalabnormalities,andreinforcestheneedformoreresearchintothemechanismsunderlyingthisrelationship.Giventhehighprevalenceofobesityandmentalhealthproblemsinmanypopulations,thesefindingshaveimportantimplicationsforpublichealthandhighlighttheurgentneedfornewapproachestopreventionandtreatmentInadditiontotheimplicationsforpublichealth,thefindingsofthisstudyalsohaveimportantimplicationsforclinicalpractice.Healthcareprovidersshouldrecognizethepotentialimpactofdiet-inducedobesityonmentalhealthandconsidertheimplementationofinterventionsthatfocusonbothphysicalandmentalwellbeing.

Forexample,interventionsthataddressbothdietandexercisecouldhavesignificantbenefitsforpatientswithobesityandmentalhealthconditions.Theseinterventionscouldincludestructuredexerciseprograms,dietarycounseling,andbehavioraltherapiesthataddresstheunderlyingcausesofunhealthyeatinghabitsandprovidepatientswiththeskillsandtoolsnecessarytomakesustainablelifestylechanges.

Furthermore,interventionsthatspecificallytargetthemicrobiomecouldalsobebeneficial.Asresearchintothegut-brainaxiscontinuestoevolve,itisbecomingincreasinglyclearthatthemicrobiomeplaysacriticalroleinregulatingmoodandbehavior.Therefore,interventionsthatmodulatethecompositionandfunctionofthemicrobiomecouldhavesignificantimplicationsformentalhealth.

Inconclusion,therelationshipbetweendiet-inducedobesityandemotionalabnormalitiesisacomplexandmultifacetedissuethatrequiresfurtherinvestigation.Thefindingsofthisstudyunderscoretheimportanceofaddressingbothphysicalandmentalhealthinthepreventionandtreatmentofobesityandrelatedmentalhealthcon
