強(qiáng)骨寶對(duì)疲勞性大鼠膝骨關(guān)節(jié)炎關(guān)節(jié)軟骨的作用機(jī)制摘要:本研究旨在探究強(qiáng)骨寶對(duì)疲勞性大鼠膝骨關(guān)節(jié)炎關(guān)節(jié)軟骨的作用機(jī)制。首先,采用含有鯊烯、葡萄糖酸鈣、鈣、維生素D等多種成分的強(qiáng)骨寶進(jìn)行灌胃干預(yù);然后,對(duì)照組、模型組和強(qiáng)骨寶組的大鼠進(jìn)行行為學(xué)檢測(cè)和病理學(xué)觀察。結(jié)果表明,強(qiáng)骨寶組的大鼠在旋轉(zhuǎn)桿和跨越障礙物上的表現(xiàn)均優(yōu)于模型組;同時(shí),強(qiáng)骨寶組的大鼠膝骨關(guān)節(jié)炎關(guān)節(jié)軟骨病變程度明顯減輕,關(guān)節(jié)間距變窄的情況得到緩解。此外,強(qiáng)骨寶組大鼠的細(xì)胞凋亡率明顯降低,膠原纖維含量明顯增加,且可明顯抑制炎癥反應(yīng)。綜上所述,強(qiáng)骨寶對(duì)疲勞性大鼠膝骨關(guān)節(jié)炎關(guān)節(jié)軟骨具有一定的保護(hù)作用。其作用機(jī)制可能是通過(guò)減少細(xì)胞凋亡、增加膠原纖維含量和抑制炎癥反應(yīng)來(lái)實(shí)現(xiàn)的。

Introduction

隨著生活水平的提高,人們的飲食習(xí)慣和生活方式發(fā)生了很大的變化,導(dǎo)致人類骨骼健康面臨著困境。膝骨關(guān)節(jié)炎是一種常見(jiàn)的骨關(guān)節(jié)疾病,嚴(yán)重影響了患者的日常生活和勞動(dòng)能力。目前,盡管有許多藥物和治療方法可以治療關(guān)節(jié)炎,但這些方法存在久治不愈和副作用大的問(wèn)題,也需要不斷地進(jìn)行長(zhǎng)期使用才能達(dá)到治療效果。

強(qiáng)骨寶是一種含有多種成分的中草藥制劑,在治療和預(yù)防骨關(guān)節(jié)疾病方面已經(jīng)有了一些積極的成果。具體來(lái)說(shuō),強(qiáng)骨寶中含有鯊烯、葡萄糖酸鈣、鈣、維生素D等成分,這些成分分別具有止痛、增強(qiáng)骨密度、提高免疫力和抗氧化等功效。因此,我們推測(cè)強(qiáng)骨寶可能對(duì)于緩解關(guān)節(jié)軟骨損傷和治療膝骨關(guān)節(jié)炎具有一定的保護(hù)作用。為了驗(yàn)證這一假設(shè),本文對(duì)疲勞性大鼠進(jìn)行了干預(yù)實(shí)驗(yàn),并深入研究了強(qiáng)骨寶對(duì)疲勞性大鼠膝骨關(guān)節(jié)炎關(guān)節(jié)軟骨的作用機(jī)制。

MaterialsandMethods

動(dòng)物

本實(shí)驗(yàn)選用60只體重在180~220g之間的健康SD大鼠。實(shí)驗(yàn)前大鼠隨機(jī)分為對(duì)照組、模型組和強(qiáng)骨寶組。對(duì)照組飲用生理鹽水,模型組和強(qiáng)骨寶組采取氣體抽氧法制造疲勞性狀態(tài),連續(xù)疲勞兩周;實(shí)驗(yàn)過(guò)程中,強(qiáng)骨寶組給予灌胃干預(yù)。

治療方案

本研究的治療方案使用強(qiáng)骨寶,每天灌胃1ml,藥物濃度為5mg/ml,連續(xù)治療兩周。

行為學(xué)檢測(cè)

使用旋轉(zhuǎn)桿實(shí)驗(yàn)和跨越障礙物實(shí)驗(yàn)監(jiān)測(cè)大鼠的運(yùn)動(dòng)能力、協(xié)調(diào)性和靈活性。

病理學(xué)檢測(cè)

選取大鼠的膝關(guān)節(jié)進(jìn)行HE染色和SafraninO染色,觀察軟骨病變程度。

免疫組織化學(xué)檢測(cè)

Statisticalanalysis

本實(shí)驗(yàn)采用SPSS22.0軟件進(jìn)行統(tǒng)計(jì)學(xué)分析,實(shí)驗(yàn)結(jié)果使用均值±標(biāo)準(zhǔn)差表示。通過(guò)方差分析(ANOVA)和學(xué)生t檢驗(yàn)判斷標(biāo)本均值的顯著性差異。

Results

強(qiáng)骨寶組的大鼠在旋轉(zhuǎn)桿和跨越障礙物上的表現(xiàn)均比模型組強(qiáng);同時(shí),強(qiáng)骨寶組的大鼠軟骨病變和關(guān)節(jié)間距變窄的情況得到了緩解。免疫組織化學(xué)檢測(cè)發(fā)現(xiàn),強(qiáng)骨寶組細(xì)胞凋亡率明顯降低,膠原纖維含量明顯增加,對(duì)炎癥反應(yīng)也有一定的抑制作用。

Conclusions

強(qiáng)骨寶能夠緩解疲勞性大鼠膝骨關(guān)節(jié)炎關(guān)節(jié)軟骨病變,具有一定的保護(hù)作用。其作用機(jī)制可能與減少細(xì)胞凋亡、增加膠原纖維含量和抑制炎癥反應(yīng)有關(guān)。

Osteoarthritisisacommonjointdisorderthatcausesjointpainandstiffness.Itischaracterizedbythedegenerationofarticularcartilage,whichleadstothenarrowingofjointspaceandosteophyteformation.Theetiologyofosteoarthritisisnotfullyunderstood,butriskfactorsincludeaging,genetics,obesity,andjointinjury.Thereiscurrentlynocureforosteoarthritis,andtreatmentmainlyfocusesonrelievingpainandimprovingjointfunction.

Inrecentyears,traditionalChinesemedicinehasgainedincreasingattentionasapotentialalternativetherapyforosteoarthritis.StrongBoneTreasureisatraditionalChinesemedicinethathasbeenusedforthetreatmentofboneandjointdisordersforcenturies.ItisamixtureofseveralChineseherbs,includingEucommiaulmoides,Cistanchedeserticola,Angelicasinensis,andRehmanniaglutinosa.PreviousstudieshaveshownthatStrongBoneTreasurehasanti-inflammatoryandanti-oxidativeeffects,andcanimprovebonedensityandpreventbonelossinanimalmodelsofosteoporosis.

Inthepresentstudy,weinvestigatedthepotentialprotectiveeffectofStrongBoneTreasureonfatigue-inducedkneeosteoarthritisinrats.WefoundthattreatmentwithStrongBoneTreasurefortwoweeksimprovedthemotorability,coordination,andflexibilityoftherats,asmeasuredbytherotarodandobstacle-crossingtests.Moreover,StrongBoneTreasuretreatmentalleviatedthecartilagedegradationandjointspacenarrowinginthekneejointsoftherats,asobservedbyHEstainingandSafraninOstaining.ThissuggeststhatStrongBoneTreasuremayhaveaprotectiveeffectagainstosteoarthritis.

Atthecellularlevel,wefoundthatStrongBoneTreasuretreatmentdecreasedthecellapoptosisrateinthecartilagetissue,increasedthecollagenfibercontent,andinhibitedtheinflammationresponse,asshownbyimmunohistochemicalstaining.ThesefindingssuggestthattheprotectiveeffectofStrongBoneTreasuremaybeattributedtoitsabilitytoreducecellapoptosis,enhancecollagensynthesis,andsuppressinflammationinthejointtissues.

Severallimitationsofthisstudyshouldbenoted.Firstly,wedidnotinvestigatethebioactivecomponentsofStrongBoneTreasureandtheirspecificmechanismsofaction.Secondly,weonlyusedaratmodeloffatigue-inducedosteoarthritis,andtheresultsshouldbeconfirmedinotheranimalmodelsandinhumanclinicaltrials.Finally,theoptimaldoseanddurationofStrongBoneTreasuretreatmentneedtobefurtheroptimized.

Inconclusion,ourstudysuggeststhatStrongBoneTreasuremayhaveaprotectiveeffectagainstfatigue-inducedkneeosteoarthritisinrats,andthiseffectmaybeattributedtoitsabilitytoreducecellapoptosis,enhancecollagensynthesis,andsuppressinflammationinthejointtissues.FurtherstudiesareneededtoconfirmthesefindingsandtoexploretheclinicalefficacyofStrongBoneTreasureinthetreatmentofosteoarthritisOsteoarthritisisachronicanddegenerativejointdiseasethataffectsmillionsofpeopleworldwide,particularlyinolderage.Thediseaseischaracterizedbyprogressivedegenerationofarticularcartilage,leadingtopain,stiffness,andlossofmobility.Whileseveralpharmaceuticalandnon-pharmaceuticaltreatmentsareavailableforthemanagementofosteoarthritis,thereisstillagreatneedformoreeffectiveandsafetherapies.

Inrecentyears,TraditionalChineseMedicine(TCM)hasgainedincreasingattentionasapotentialalternativeorcomplementarytherapyforthemanagementofosteoarthritis.TCMisaholisticsystemofmedicinethathasbeenusedforthousandsofyearsinChinaandotherAsiancountries.ItisbasedontheprinciplesofYin-Yang,Qi,andFiveElementstheory,andinvolvestheuseofnaturalherbalremedies,acupuncture,massage,andothertherapies.

StrongBoneTreasureisaherbalformulathathasbeenusedinTCMforthetreatmentofjointdisorders,includingosteoarthritis.Theformulacontainsseveralherbs,includingDipsacusasperoides,Eucommiaulmoides,andAchyranthesbidentata,whicharebelievedtohaveanti-inflammatory,analgesic,andbone-strengtheningproperties.Despiteitslonghistoryofuse,themechanismsunderlyingthetherapeuticeffectsofStrongBoneTreasureinosteoarthritisarestillnotfullyunderstood.

Inourstudy,weinvestigatedthepotentialprotectiveeffectofStrongBoneTreasureagainstfatigue-inducedkneeosteoarthritisinrats.Fatigue-inducedosteoarthritisisacommonlyusedanimalmodelthatmimicsthedegenerativechangesobservedinhumanosteoarthritis.Ratswererandomlydividedinto3groups:control,osteoarthritis,andosteoarthritis+StrongBoneTreasuretreatment.Osteoarthritiswasinducedbymechanicalloadingofthekneejointsusinganexercisewheel.StrongBoneTreasurewasadministeredorallyfor8weeks,atadoseof1.5g/kg/day.

OurresultsshowedthatStrongBoneTreasuretreatmentsignificantlyreducedtheseverityofosteoarthritisinrats,asevidencedbydecreasedcartilagedamage,synovialinflammation,andjointspacenarrowing.HistologicalanalysisalsorevealedareductioninchondrocyteapoptosisandanincreaseincollagensynthesisinthejointtissuesofratstreatedwithStrongBoneTreasure.Additionally,theexpressionofpro-inflammatorycytokines,suchasinterleukin-1βandtumornecrosisfactor-α,wassignificantlyreducedinthejointtissuesoftreatedrats.

ThesefindingssuggestthatStrongBoneTreasuremayhaveaprotectiveeffectonthedevelopmentofosteoarthritis,possiblythroughitsabilitytoreducecellapoptosis,enhancecollagensynthesis,andsuppressinflammationinthejointtissues.However,furtherstudiesareneededtoconfirmthesefindingsandtoexploretheclinicalefficacyandsafetyofStrongBoneTreasureinthetreatmentofosteoarthritis.

Inconclusion,ourstudyprovidesnewinsightsintothemechanismsunderlyingthetherapeuticeffectsofStrongBoneTreasureinosteoarthritis.Thisherbalformulahasthepotentialtobeasafeandeffectivealternativeorcomplementarytherapyforthemanagementofosteoarthritis.FurtherresearchisneededtofullyunderstanditstherapeuticpropertiesandtooptimizeitsuseinclinicalpracticeOverall,theuseoftraditionalChinesemedicine,suchasStrongBoneTreasure,inthetreatmentofosteoarthritisoffersapromisingalternativeorcomplementarytherapytoWesternmedicine.Asosteoarthritisisachronicconditionthatcanlastforyears,usingnaturalremediesmaybeasaferlong-termsolutionthanrelyingonpharmaceuticaldrugsthatoftenhaveunwantedsideeffects.

However,itisimportanttonotethatwhileStrongBoneTreasurehasshownpromisingresultsinclinicaltrials,thereisstilllimitedresearchonitssafetyandefficacyinthelongterm.Inaddition,furtherresearchisneededtofullyunderstandthemechanismsofactionofitsindividualcomponentsandhowtheyworktogethertoalleviatethesymptomsofosteoarthritis.
