摘要:牙周疾病是一種常見(jiàn)的口腔疾病,其主要導(dǎo)致牙周組織的破壞和牙齒的喪失。干細(xì)胞治療成為牙周疾病的一種新興治療方法。本研究旨在探究液態(tài)濃縮生長(zhǎng)因子對(duì)人牙周膜干細(xì)胞成骨能力的影響。通過(guò)對(duì)人牙周膜干細(xì)胞進(jìn)行培養(yǎng),加入不同濃度的液態(tài)濃縮生長(zhǎng)因子,觀察其對(duì)干細(xì)胞的生長(zhǎng)、增殖和成骨能力的影響。結(jié)果表明,不同濃度的液態(tài)濃縮生長(zhǎng)因子對(duì)人牙周膜干細(xì)胞的成骨能力有顯著的影響。適當(dāng)?shù)臐舛纫簯B(tài)濃縮生長(zhǎng)因子可以提高干細(xì)胞成骨能力,增加新骨生成率。本研究對(duì)引導(dǎo)牙周膜干細(xì)胞成骨治療提供理論基礎(chǔ)。

Abstract:Periodontaldiseaseisacommonoraldisease,whichmainlyleadstothedestructionofperiodontaltissuesandthelossofteeth.Stemcelltherapyhasbecomeanemergingtreatmentforperiodontaldisease.Thepurposeofthisstudywastoinvestigatetheeffectofliquidconcentratedgrowthfactorsontheosteogenicabilityofhumanperiodontalligamentstemcells.Humanperiodontalligamentstemcellswereculturedanddifferentconcentrationsofliquidconcentratedgrowthfactorswereaddedtoobservetheireffectonthegrowth,proliferation,andosteogenicabilityofstemcells.Theresultsshowedthatdifferentconcentrationsofliquidconcentratedgrowthfactorshadasignificanteffectontheosteogenicabilityofhumanperiodontalligamentstemcells.Appropriateconcentrationofliquidconcentratedgrowthfactorscanimprovetheosteogenicabilityofstemcellsandincreasethenewboneformationrate.Thisstudyprovidesatheoreticalbasisforguidingperiodontalligamentstemcellstobecomeosteogenictherapy.

Inrecentyears,concentratedgrowthfactors(CGFs)havebeenusedasasourceofgrowthfactorsfortissueregeneration.CGFsarederivedfromthepatient'sownbloodandcontainahighconcentrationofgrowthfactors,includingplatelet-derivedgrowthfactor,transforminggrowthfactor-β,insulin-likegrowthfactor,andvascularendothelialgrowthfactor.Liquidconcentratedgrowthfactors(LCGFs)areanewerformofCGFs,whichareobtainedbycentrifugingthewholebloodwithoutaddinganticoagulants.
Inthisstudy,theresearchersinvestigatedtheeffectofLCGFsontheosteogenicabilityofhPDLSCs.TheresultsshowedthatLCGFssignificantlyimprovedtheosteogenicdifferentiationofhPDLSCsinaconcentration-dependentmanner.TheoptimalconcentrationofLCGFswasfoundtobe10%(v/v).Moreover,LCGFsincreasedtheexpressionofosteogenicmarkers,suchasalkalinephosphatase,osteopontin,andosteocalcin,andenhancedthemineralizationofextracellularmatrix.
ThesefindingssuggestthatLCGFscanenhancetheosteogenicabilityofhPDLSCsandpromoteboneformation.LCGFsmayhavepotentialapplicationsinthefieldofperiodontaltissueregeneration.However,furtherstudiesareneededtoinvestigatethelong-termeffectsandsafetyofLCGFsinclinicalsettingsInadditiontothepotentialapplicationsinperiodontaltissueregeneration,LCGFsmayalsohaveimplicationsinotherfieldsoftissueengineeringandregenerativemedicine.Forinstance,LCGFsmayserveasanalternativeorcomplementaryapproachtotraditionalgrowthfactorsinpromotingbonehealingandregenerationinorthopedicsurgery.
Furthermore,theuseofLCGFsmayalsohaveadvantagesintermsofcost-effectivenessandsafety.Traditionalgrowthfactorscanbeexpensiveandmayhaveadverseeffects,suchasinflammationandtumorformation,whichlimittheirclinicalapplication.LCGFs,ontheotherhand,arederivedfromautologousbloodandarethereforeinexpensiveandsafe.
Nevertheless,therearestillseveralchallengesthatneedtobeaddressedbeforetheclinicaltranslationofLCGFs.OnechallengeisthestandardizationofthepreparationandadministrationprotocolofLCGFs.Currently,thereisalackofconsensusontheoptimalconcentrationandvolumeofLCGFstobeusedindifferentclinicalscenarios.Therefore,standardizationoftheprotocolisessentialtoensuretheefficacyandsafetyofLCGFs.
AnotherchallengeistheidentificationoftheoptimalsourceofLCGFs.Asmentionedearlier,theconcentrationsofgrowthfactorsinbloodcanvarydependingonthedonor'sage,healthstatus,andotherfactors.Therefore,theidentificationoftheoptimalsourceofbloodandthestandardizationofthecollectionandprocessingprotocolarecrucialforthesuccessfulapplicationofLCGFsinclinicalsettings.
Overall,LCGFsshowgreatpromiseinpromotingtissueregenerationinvariousclinicalscenarios.Furtherstudiesareneededtoinvestigatethelong-termeffectsandsafetyofLCGFsinclinicalsettingsandtooptimizetheprotocolfortheirpreparationandapplication.Withfurtherresearch,LCGFsmaybecomeavaluabletoolfortissueengineeringandregenerativemedicineInadditiontotheiruseintissueengineeringandregenerativemedicine,LCGFshavealsoshownpotentialinotherclinicalapplications.Forexample,arecentstudydemonstratedtheefficacyofLCGFsinreducingpainandpromotinghealinginpatientswithdiabeticfootulcers.TheLCGFswerepreparedfromthepatient'sownbloodandapplieddirectlytotheulcersite,resultinginsignificantreductionsinpainandimprovementsinhealing.
LCGFshavealsobeeninvestigatedasapotentialtreatmentforosteoarthritis.Inapreclinicalstudy,LCGFswereinjectedintothekneejointsofmicewithinducedosteoarthritis,leadingtoincreasedcartilageregenerationandimprovementsinjointfunction.Additionally,aclinicaltrialiscurrentlyunderwaytoinvestigatethesafetyandefficacyofLCGFsintreatingkneeosteoarthritisinhumans.
AnotherpotentialapplicationofLCGFsisinthetreatmentofhairloss.SeveralstudieshavedemonstratedtheabilityofLCGFstopromotehairgrowthandincreasehairdensityinpatientswithandrogeneticalopecia.TheLCGFswerepreparedfromthepatient'sownbloodandinjectedintothescalp,resultinginsignificantimprovementsinhairgrowthandthickness.
Overall,LCGFsshowgreatpromiseinawiderangeofclinicalapplications.However,furtherresearchisneededtofullyunderstandtheirmechanismofaction,optimizetheirpreparationandapplicationprotocols,andinvestigatetheirlong-te
