BPG層析柱標(biāo)準(zhǔn)裝柱流程規(guī)范

1、操作標(biāo)準(zhǔn)冃的：為了使bpg300層析柱裝柱過程可重復(fù)，并能夠得到較好的柱效，故建立此 操作規(guī)范。范圍：配合aktaprocess系統(tǒng)進(jìn)行有效的蛋白質(zhì)純化操作。責(zé)任人：層析工序操作人員標(biāo)準(zhǔn)操作程序：1基本術(shù)語概念填料因子（packing factor，pf）:指在在裝柱過程中，用低流速（如60cm/h）對填料進(jìn)行沉降壓縮得到的調(diào)料高度和最后裝柱完成時填料高度（柱高）的比 值，gehc生產(chǎn)的各種常見填料的pf參見gehc axichrom 300-1000 columns user manual 28-9562-89 edition aa；packing factor pf= (consolida

5、min 表示；換算:ml錢性 jisj®體和流速=6llx柱橫截面面積2準(zhǔn)備工作2.1檢查層析柱的完整性，所帶配件，是否水平（用水平儀矯正），是否清潔 （如有污染應(yīng)進(jìn)行淸洗）。2.2確定所用填料及其特性、濃度等以deae sepharose fast flow填料為例，其填料因子（pf）為1.15,新填料 （slurry）濃度為75% （保存于20%乙醇屮），耐壓3bar。2.3確定放大后的工藝參數(shù)以xx生化制藥股份有限公司的放大工藝為例，屮試工藝參數(shù)：柱高h(yuǎn)=10cm；體積流速vl=3ml/min；柱內(nèi)徑d= 1.6cm.即，線性流速 v2=vl/s 柱=3/（3.14x0.82）

6、 cm/min=1.5 cm/min；樣品與填料接觸時間t=h/v2= 10/1.5min=6.67min；在工藝的線性放大中h、v2和t均不變，則可保證樣品出峰的位置（洗脫吋間）和形 狀不變。使用bpg300層析柱進(jìn)行放大的工藝參數(shù)：裝柱體積 v 林=s tth=3.14xl.52xll=7l；裝柱需要 slurry 體積 vslurry=v 柱xpf/cslurry以 deae sepharose fast flow 為填料，則，vslurty=7x 1.15/0.75l= 10.73l；裝入柱中的 slurry 高度 ls|urry=vsiurry/s10.73/（3.14x 1.52）

7、dm= 1.52dm= 15.2cm.3管路安裝3.1層析柱管道和配件安裝3.1.1卸下柱子頂部和底部的盲堵墊圈，旋松密封調(diào)節(jié)器，用純水或20%乙醇 充分浸潤柱內(nèi)壁和柱頭o型圈；3.1.2用扳手對稱卸下支持桿連接到法蘭的螺絲，并通過旋轉(zhuǎn)高度調(diào)節(jié)手柄將柱 頭旋至最高處，在柱頭頂部依次安裝壓力表和四路二通閥，在柱底安裝四路二通閥；3.1.3在頂部四路二通閥上橫向連接一根細(xì)管，另一端連至akta process上端 連接口 column top,同時從柱底四路二通閥橫向接出一根細(xì)管至akta process 的卜端連接口 column bottom。圖1、bpg300層析柱安裝示意圖table 1

13、e 運 行程序充水至bpg柱中2-3cm左右，pause,清除下端篩網(wǎng)中的空氣，用注射 器吸出篩網(wǎng)中殘留的氣泡。4裝填料4.1準(zhǔn)備填料所用填料為deae sepharose fast flow ,裝置的是保存于20%乙醇巾的新 slurry；4.2卸下柱頭，用攪拌棍引流加入混勻的slurry至15.2cm處，再加入純水至 40cm左右，充分?jǐn)嚢杌靹?，同時避免擠壓篩網(wǎng)和柱壁，靜置使填料界面至少 低于38cm；4.3 在 system control 窗口點擊 manual-*valves-*inleta,選擇 inletai，insert； inletb,選擇 closed, insert； c

14、olumn,選擇 downflow，insert； pump-*flow,設(shè)定流速60l/h,點擊execute運行程序?qū)⒅^上端篩網(wǎng)充滿水以除去氣泡，再 裝上柱頭，用密封調(diào)節(jié)器固定柱尖，擰緊螺絲；4.4 在 system control 窗 口點擊 manual-*pump-*flow,設(shè)定流速為 42l/h (60cnvh),進(jìn)行填料的沉降，待填料高度降至14cm左右，將密封調(diào)節(jié)器旋至全松，將柱頭快速調(diào)至距離填料3-4cm處，將密封調(diào)節(jié)器旋至半松，將柱頭快 速調(diào)至濾網(wǎng)距離填料l-2cm,旋緊密封調(diào)節(jié)器，再設(shè)定高流速(如150l/h),繼續(xù)壓縮填料，最后再調(diào)節(jié)柱頭至10cm。整個過程一直有流

15、速，泵速不能停。 5柱效測定圖2、柱效測定示意圖5.1平衡將a1接口細(xì)管放入0.4m nacl溶液中，將柱頂端四路二通閥調(diào)至管路與大 氣相連狀態(tài)，在 system control 窗口點擊 manual -* valves-* inlet a,選擇 inletal， insert； inletb,選擇 closed，insert； column,選擇 downflow, insert； pump-* flow,再設(shè)定高流速（如150l/h），點擊execute運行，排氣泡，再將閥調(diào)至管 路與柱頭相連狀態(tài)，平衡3-5個柱體積至電導(dǎo)曲線穩(wěn)定，點擊end:5.2上樣5.2.1將b1接u細(xì)管放入2m

16、nacl溶液，柱頂端四路二通閥調(diào)至管路與大氣相 連狀態(tài)，在 system control 窗口點擊 manualvalvesinleta，選擇 closed, insert； inletb,選擇 inletb 1, insert； column,選擇 downflow, insert； pump -flow，設(shè)定高流速（150l/h）點擊execute運行，排氣泡，pause；5.2.2 在 system control 窗 u 點擊 manualother-*end_timer,選擇 acc.volumn, timeout設(shè)上樣量力70ml （1 %柱體積）；pumpflow,設(shè)定上樣流 速

17、60l/h,點擊execute的同時將柱頂端四路二通閥調(diào)至管路與柱頭相連，完 成上樣；column packing檢測裝柱效果一般用<0.5% （30um介質(zhì)）或 <1% （90um介質(zhì)）柱體積的1%丙酮測柱效和 峰形 the linear flow rate should be 30 cm/h for 34 pm and 50 pm media and 20 cm/h for 90 pm media5.3洗脫5.3.1將柱頂端四路通閥調(diào)至管路與大氣相連狀態(tài)，在system control窗口點 擊 manual-*valves-*inleta，選擇 inletal, insert

18、； inletb,選擇 closed，insert； column，選擇 downflow，insert； pump-*flow,設(shè)定高流速(150l/h)點擊 execute運行，排氣泡，同時將管路充滿0.4m nacl溶液，調(diào)流速至21l/h(30cm/h);5.3.2 待流速穩(wěn)定后，在 system control 窗 口點擊 manual-*valves-injection mark, insert,點擊execute的同吋將柱頂端四路二通閥調(diào)至管路與柱頭相連， 開始洗脫，在一個柱體積左右，電導(dǎo)檢測下將產(chǎn)生目的峰。5.4測柱效在evaluation窗口屮，選擇實驗保存的結(jié)果，在結(jié)果圖屮

19、點擊鼠標(biāo)右鍵，選 擇 properties,點擊 clear,并選中 cond_102，點擊 0k?；氐?evaluation 窗口, 選擇釈分按鈕，在彈出窗口中，選擇cond_102，peak window中調(diào)整積分的范 圍，并在 reject peaks 中，填 peak must be one of 1 largest；確定后，在積分結(jié) 果窗口，點擊右鍵，選擇properties，在peak table選項屮，勾選plate height(hetp)和asymmetry。確定后即可計算得到計算理論塔板高度hetp和對 稱因子as。appendix b packing buffer inf
